For that iconic Spaghetti Western sound, take an electric guitar with a single-coil pickup at the bridge and plug it into a Fender amp with Reverb to max. Make an E-minor chord (or B-minor, if using the baritone guitar shown) with a single upward strum close to the bridge, and let it ring.
(A dreadnought acoustic guitar will do, in a pinch.)
Cue the "Red-tail Hawk" SFX.
